چکیده

The aim of this study is to optimize the production of Baker’s yeast, Citric acid, Ethanol and α-amylase using dates of low market value as the basal fermentation medium. The results of this study showed that the optimal Baker’s yeast production was obtained with a dilution rate of 0.22 h-1. Also, the use of ammonium phosphate provides better biomass yields compared to urea. As for the Ethanol production, the optimum yield obtained was 131.0 g/L under optimum conditions of an incubation period of 72 h, inoculums content of 4 % (w/v), sugars concentration of 180.0 g/L and ammonium phosphate concentration of 1.0 g/L. Concerning the Citric acid production, the cumulative effect of temperature (30 °C), methanol concentration (3%), initial pH (3.5) and ammonium nitrate concentration (2.5 g/L) during the fermentation process of dates syrup did increase the citric acid production to 97.0 g/L. Finally, the maximum α-amylase activity obtained was 2600 μmol/L/min under optimum conditions of an incubation period of 96 h, temperature of 30 °C, an initial pH of 5.5, sugars concentration of 20 g/L, yeast extract concentration of 5.0 g/L and phosphorus concentration of 5.0 g/L. Keys words: Dates/ Baker’s yeast / Ethanol/ Citric acid/ α-amylase

Process optimization for ethanol production from very high gravity (VHG) finger millet medium using response surface methodology

The Box-Wilson central composite design (CCD) based on response surface methodology (RSM) was used for ethanol fermentation using very high gravity (VHG) finger millet hydrolysate. Optimized process variables were namely, concentrations of yeast extract, magnesium sulphate and pH of the medium. High gravity mashes (>300 g dissolved solids per liter) were prepared by a thermo-stable α-amylase, f...

Production ofHalothermotolerant α-Amylase from aModerately Halophilic Bacterium, NesterenkoniaStrain F.

Production of extracellular amylase was demonstrated under conditions of high salinity in aerobically cultivated culture of a newly isolated moderately halophilic Gram-positive coccus, designated strain F in basal medium containing peptone from meat, yeast extract, NaCl (7% w/v) and starch. Biochemical and physiological characterization along with 16S rRNA sequence analysis placed F in the genu...

Utilization of date syrup as a substrate for carotenoid production by Rhodotorula glutinis

The potential use of date syrup, for the production of carotenoids by Rhodotorula glutinisin batch fermentation process, was investigated during 7 days. The results revealed that carbon (glucose or date syrup) and nitrogen sources [yeast extract, (NH4)2SO4 and NH4NO3] had a significant influence on biomass and carotenoid production. Maximum yield of total carotenoid production (7.94 mg/L) with ...

Efficient Expression of Maltohexaose-Forming α-Amylase from Bacillus stearothermophilus in Brevibacillus choshinensis SP3 and Its Use in Maltose Production

The maltohexaose-forming, Ca2+-independent α-amylase gene from Bacillus stearothermophilus (AmyMH) was efficiently expressed in Brevibacillus choshinensis SP3. To improve the production of AmyMH in B. choshinensis SP3, the temperature and initial pH of culture medium were optimized. In addition, single-factor and response surface methodologies were pursued to optimize culture medium. Addition o...

A novel high ethanol-thermo-tolerant Acetobacter pasteurianus KBMNS-IAUF-2 strain and the optimization of acetic acid production using the Taguchi statistical method

Because of the high energy consumption for fermentor cooling, the isolation of thermo-tolerant Acetobacter strains for vinegar production has a high priority. The aims of this study were the isolation and identification of a high ethanol-thermo-tolerant Acetobacter spp. from grapes as well as the optimization of conditions for increasing the acetic acid production. The grape e...
